Phenomenological modeling of DNA overstretching
A phenomenological model based on the three-dimensional theory of nonlinear elasticity is developed to describe the phenomenon of overstretching in the force-extension curve for dsDNA. By using the concept of a material with multiple reference configurations a single formula is obtained to fit the force-extension curve.
متن کاملCoarse-grained simulations of DNA overstretching.
We use a recently developed coarse-grained model to simulate the overstretching of duplex DNA. Overstretching at 23 °C occurs at 74 pN in the model, about 6-7 pN higher than the experimental value at equivalent salt conditions. Furthermore, the model reproduces the temperature dependence of the overstretching force well. The mechanism of overstretching is always force-induced melting by unpeeli...
متن کاملPeeling back the mystery of DNA overstretching.
W hen a single DNA molecule is stretched beyond its normal B-form contour length, it will undergo one of two types of conformational transitions at high or low force, depending on its attachment and ability to rotate. The structure of the DNA during these transitions is difficult to measure with traditional structural methods because it only occurs on a single molecule. More than a decade after...
متن کاملDNA overstretching transition: ionic strength effects.
As double-stranded DNA is stretched to its B-form contour length, models of polymer elasticity can describe the dramatic increase in measured force. When the molecule is stretched beyond the contour length, it further shows a highly cooperative overstretching transition. We have developed a theoretical description for this transition by coupling the two-state model and the elasticity theory pro...
متن کاملTheory of high-force DNA stretching and overstretching.
Single-molecule experiments on single- and double-stranded DNA have sparked a renewed interest in the force versus extension of polymers. The extensible freely jointed chain (FJC) model is frequently invoked to explain the observed behavior of single-stranded DNA, but this model does not satisfactorily describe recent high-force stretching data. We instead propose a model (the discrete persiste...
